Gene Expression Caused by Alkylating Agents and cis- Diamminedichloroplatinum(II) in Escherìchiacoli1

چکیده
Previous work has demonstrated heterogeneous effects of methylating agents on induction of DNA damage inducible genes in Escherìchia coli. These studies employed /:'. coli mutants that have fusions of the lac operon to genes induced by treatment with sublethal levels of alkylating agents. These mutants were selected from random insertions of the Mu di (Ap1lac) phage by screening for induction of /3-galactosidase activity in the presence of methylmethanesulfonate or W-methyl-W-nitro-Nnitrosoguanidine.The current report extends these findings by analyzing gene expression caused by mechlorethamine, chloroethylnitrosoureasand ciVdiamminedichloroplatinum(II) (m-DDP). The results demonstrate heterogeneous effects by these agents on gene expression. While l-(2chloroethyi)-3-cyclohexyI-l-nitrosourea induces ulkA, other nitrosoureas, mechlorethamine, and cis-DDP do not cause expression of this gene. Further, while all nitrosoureas caused expression ofaidC, mechlorethamine and ciVDDP did not. Lastly, c/'.v-l)DPcaused marked expres sion of a su!A fusion mutant while not inducing any of the other /•". coli

Gene expression caused by alkylating agents and cis-diamminedichloroplatinum(II) in Escherichia coli.
Previous work has demonstrated heterogeneous effects of methylating agents on induction of DNA damage inducible genes in Escherichia coli. These studies employed E. coli mutants that have fusions of the lac operon to genes induced by treatment with sublethal levels of alkylating agents. These mutants were selected from random insertions of the Mu-dl (Apr lac) phage by screening for induction of...
متن کاملEffect of novobiocin on the antitumor activity and tumor cell and bone marrow survivals of three alkylating agents.
Our previous in vitro studies demonstrated marked synergy with alkylating agents when novobiocin was present during and after alkylating agent exposure. To determine whether this effect is observed in vivo, novobiocin was administered daily for 3 days prior to alkylating agent treatment, during alkylating agent treatment, and for 2 days after completion of alkylating agent treatment. When combi...

Previous studies of ERCC-1 gene expression levels in chronic lymphocytic leukemia and ovarian carcinoma tumor specimens indicated that increased gene expression may correlate with a lack of response to the alkylating agents, melphalan, and cu-diamminedichloroplatinum (II). In order to demonstrate direct involvement of the ERCC-1 protein in repair of melphalan lesions, the ERCC-1 defective Chine...
متن کاملA Novel Role for DNA Photolyase: Binding
DNA photolyase binds to and repairs cyclobutane pyrimidine dimers induced by UV radiation. Here we demonstrate that ih the yeast Saccharomyces cerevisiae, photolyase also binds to DNA damaged by the anticancer drugs cis-diamminedichloroplatinum (cis-DDP) and nitrogen mustard (HN2) and by the alkylating agent N-methyl-N'-ditro-N-nitrosoguanidine (MNNG). Surprisingly, mutations in photolyase were...
